Separation

cells

impurities
from the output of fermentation in
alkaloid
drugs
bioreactor
by
ultrafiltration
Background:
Alkaloid
drugs
are a wide organic compound which are made by living organisms. Alkaloids have at least nitrogen atoms in its structure and have a wide range of phrenological effects
such
as antibacterial and antimalaria. Nowadays, a myriad of
research
focuses on the industrial fermentative
production
of
alkaloid
drugs
from natural sources.
However
, it is relatively easy to ferment the sources, the low concentration of desired compounds makes drug discovery from fermentation
bioreactor
difficult.
Therefore
, others
research
concentrates on downstream processing which is costly part in bio-
drugs
production
. Downstream processing entails the recovery bio-product from natural sources
such
as
host
cells
and elimination
impurities
from complex fermentation
broth
in order to recycle of salvageable materials. The output of fermentation
bioreactor
is a mixture of diverse molecules which have different in the physical and chemical properties.
Therefore
, downstream processing typically determined by economic feasibilities of the approach.
Ultrafiltration
and microfiltration are a common step in
separation
of
host
cell and
impurities
because of high selectivity and low operation conditions. The
ultrafiltration
membrane
can be combined by fermentation
bioreactor
so as to increase the productivity of the fermentation.
Ultrafiltration
is a
membrane
separation
process
that is
used in a vast wastewater and food industries
such
as dairy.
Ultrafiltration
is the most convenient
membrane
for setting the level of protein in milk and whey.
Also
, the all protein and solids in milk which have molecular weight higher than fat globules are eliminated by
ultrafiltration
membrane
.
Ultrafiltration
are be installed before solvent extraction in order to increase solvent extraction efficiency by avoiding emulsification Li et al., 2004.
Moreover
, the two-step
ultrafiltration
is used in purification of benzylpenicillin from fermented
broth
by Nabais and Cardoso (1999).
This
research
evaluated three
ultrafiltration
with molecular weight cut-off (
MWCO
) 8000,20000 and 10000. The result show high benzylpenicillin recovery and elimination proteins and
impurities
. Researchers have reported
ultrafiltration
of hemicellulose hydrolysate fermentation
broth
for xylitol
production
which is food additive and sugar substitutes. The
ultrafiltration
membrane
with 10000
MWCO

research
is part of develop a method of
alkaloid
drugs
production
in Australia company, that focuses on utilization of
ultrafiltration
for
separation
of
host
cells
and
impurities
from fermentation
broth
in downstream
production
. Methodology: The objects of these
research
are to study the performance of
ultrafiltration
on
host
cell and
impurities
separation
and to evaluate the effects of
membrane
MWCO
, transmembrane pressure, cross-flow velocity on permeate flux. 1.The experiments will conduct on the following step: 2.Preparing a fermentation
broth
of
alkaloid
bioreactor
from Australian company. 3.Design and install the
ultrafiltration
membrane
system includes recirculation systems,
ultrafiltration
modules and online permeate weight measuring. 4.Preparing and characterizing the
ultrafiltration
membrane
with a different ingredient and
MWCO
. 5.Conducting the experiment and collecting result from diverse condition. 6.Analyzing the experiment results and calculating the permeate flux and the retention ratio. Expected outcome:
This
research
will carry out to establish the highest performance of
ultrafiltration
membrane
for
separation
host
cells
and
impurities
from output of
alkaloid
fermentation
bioreactor
in optimal operation conditions.
Moreover
, the desired
ultrafiltration
membrane
will be evaluated in other operation condition and fouling of
membrane
. It is expected that productivity of
alkaloid
drugs
increases with the results of
this
research
.
